Scientists have uncovered a surprising role for a long noncoding RNA in controlling how senescent cells manage energy and stress.
The Discovery:
Researchers identified sin-lncRNA, a noncoding RNA that becomes highly active in senescent cells. By binding to the enzyme DLST, it keeps mitochondria working and prevents runaway oxidative stress. Without it, cells shift metabolism, increase damage, and reinforce aging traits.
The Science:
sin-lncRNA is induced by C/EBPβ during senescence and localizes to mitochondria.
It anchors DLST, a key enzyme of the TCA cycle, preventing its relocation to the nucleus.
Loss of sin-lncRNA impairs oxidative phosphorylation, lowers oxygen consumption, and forces cells toward glycolysis.
This shift raises reactive oxygen species (ROS), reinforcing the senescent state.
In ovarian cancer cells treated with cisplatin, sin-lncRNA rises; its depletion reduces mitochondrial activity and makes tumors more drug-sensitive.
