New transcriptomic analysis reveals that male and female aging follow entirely different biological timelines, starting years apart.
The Discovery:
By analyzing over 17,000 human transcriptomes across 35 tissues, scientists found that men experience earlier and faster molecular aging than women. These differences are most striking in the brain, where aging-related RNA splicing shifts in men strongly overlap with Alzheimer’s disease patterns. In contrast, women show delayed aging linked to hormonal regulation and mitochondrial protection.
The Science:
Transcriptome data from the GTEx project showed widespread sex differences in gene expression and alternative splicing
Male aging breakpoints appeared earlier, often between ages 35 and 50, with faster rates of transcriptomic change
Alzheimer’s-related splicing events significantly overlapped with age-associated changes in males but not females
In females, key splicing factors like SRSF1 and SRSF7 declined with age and were regulated by the estrogen receptor ESR1
Over 70% of aging-modulated genes (AMGs) were sex-specific, regulated by androgen receptors in males and estrogen receptors in females
Functional patterns diverged: males showed aging shifts in immune pathways, while females showed changes in mitochondrial and oxidative functions
