A groundbreaking study provides the first-ever senescence atlas of human skeletal muscle, revealing how aging cells drive muscle decline. This deep dive into the mechanisms of aging has led to the discovery of a new drug that acts as a potent senotherapeutic to treat age-related muscle loss.
The Discovery:
By profiling senescent cells in human muscle, scientists identified the drug Maraviroc as a potential therapeutic for sarcopenia. The study demonstrated that Maraviroc works by suppressing the harmful senescence-associated secretory phenotype (SASP), which is the major driver of inflammation and tissue disruption in aging muscles.
The Science:
The study created the first senescence atlas of human muscle, which demonstrated the widespread presence of senescent cells and their disruptive SASPs.
Maraviroc was identified as a senomorphic that blocks the Ccr5 pathway, effectively inhibiting the action of specific SASP factors that drive inflammation and aging.
In aged mice, Maraviroc treatment led to a significant increase in muscle mass and grip strength, and a pronounced reduction in cellular senescence.
The findings highlight a novel pharmacological intervention that addresses not just the symptoms of sarcopenia, but the underlying process of cellular aging itself.
