A new study explores the potential of a single gene therapy treatment to fight aging on multiple fronts. By increasing levels of the Klotho protein, researchers have successfully extended both the lifespan and healthspan of mice.

Scientists delivered a gene therapy vector coding for the secreted Klotho (s-KL) protein to wild-type mice. This single treatment resulted in a statistically significant 20% increase in lifespan in male mice. It also improved physical fitness, bone health, and neurological function.

  • The s-KL protein was delivered using an adeno-associated virus (AAV9) vector, which efficiently increased s-KL concentration in the serum.

  • Treated mice showed improved physical capacities, which was linked to a reduction in muscle fibrosis and increased muscular regenerative capacity.

  • The treatment improved bone microstructural parameters, an effect associated with osteoporosis.

  • s-KL-treated mice also exhibited increased cellular markers of adult neurogenesis and immune response in the aged hippocampus.

  • Transcriptomic analysis revealed increased phagocytosis and immune cell activity in the aged hippocampus of treated mice, without a corresponding increase in pro-inflammatory signals.
