Researchers from Hong Kong Baptist University have uncovered that inhibiting a single brain enzyme, MT1-MMP, can restore memory and learning abilities lost with age and obesity.
The Discovery:
The study demonstrates that MT1-MMP, an enzyme that increases with aging and metabolic dysfunction, directly drives cognitive decline. Blocking MT1-MMP in aged and obese mice rejuvenated hippocampal function, improved learning and memory, and restored youthful brain plasticity, without affecting neuroinflammation.
The Science:
MT1-MMP activation rises in the hippocampus during aging and correlates with neuroinflammatory markers in both mice and humans.
Genetic or pharmacological MT1-MMP inhibition (using the brain-penetrant inhibitor Ro 28-2653) restored memory and learning in aged mice and improved metabolism.
Blocking MT1-MMP in the hippocampus of obese mice reversed cognitive decline without altering inflammation.
Mechanism: MT1-MMP cleaves GPR158, a receptor critical for osteocalcin (OCN) signaling. This OCN–GPR158 axis maintains cognitive integrity.
When MT1-MMP is blocked, GPR158 remains intact, restoring OCN signaling and enhancing cognition.
Overexpression of MT1-MMP in young mice, even without inflammation, induced premature memory loss, confirming MT1-MMP alone can drive aging-like decline.

Support brain metabolism and reduce neuroinflammation naturally. Maintain healthy weight, exercise regularly, and consume polyphenol-rich foods (like berries and green tea) to help protect hippocampal function while scientists develop targeted MT1-MMP inhibitors.
