MIT researchers propose a new quantitative theory of aging, revealing that aging behaves like a dissipative system that gradually loses stability and order across cells, tissues, and genes.
The Discovery:
Scientists from the Massachusetts Institute of Technology have developed a Cellular Aging Map (CAM) that uses transformer-based AI to analyze over 65 million single-cell RNA profiles. The study shows that aging arises from dissipative dynamics, where molecular systems drift from stable (conservative) to unstable states, increasing entropy, a measurable loss of molecular order.
The Science:
Aging is modeled as a dynamical system governed by both conservative (stable) and dissipative (unstable) forces.
The team used ergodic theory and Hopf decomposition to mathematically separate these two components.
A transformer-based model trained on 1528 scRNA-seq datasets from 215 tissues and 616 cell types predicted molecular age and revealed distinct aging trajectories.
Some tissues, such as kidney and liver, showed consistent increases in entropy, signifying progressive molecular disorder.
Others, like endothelial cells, showed episodic spikes of entropy, suggesting temporary instability followed by recovery.
The model classified genes as conservative (stable, such as MKRN1, SESN1, ATR) or dissipative (variable, such as ALDH3B1, NR2C2, HERPUD1).
Disease conditions accelerated dissipation, transforming normally stable genes into unstable ones, showing pathology-driven molecular chaos.
